Implementing RLWE-based Schemes Using an RSA Co-Processor
We repurpose existing RSA/ECC co-processors for (ideal) lattice-based cryptography by exploiting the availability of fast long integer multiplication. Such co-processors are deployed in smart cards in passports and identity cards, secured microcontrollers and hardware security modules (HSM). In particular, we demonstrate an implementation of a variant of the Module-LWE-based Kyber Key Encapsulation Mechanism (KEM) that is tailored for high performance on a commercially available smart card chip (SLE 78). To benefit from the RSA/ECC co-processor we use Kronecker substitution in combination with schoolbook and Karatsuba polynomial multiplication. Moreover, we speed-up symmetric operations in our Kyber variant using the AES co-processor to implement a PRNG and a SHA-256 co-processor to realise hash functions. This allows us to execute CCA-secure Kyber768 key generation in 79.6 ms, encapsulation in 102.4 ms and decapsulation in 132.7 ms
ESCRT ubiquitin-binding domains function cooperatively during MVB cargo sorting
Ubiquitin (Ub) sorting receptors facilitate the targeting of ubiquitinated membrane proteins into multivesicular bodies (MVBs). Ub-binding domains (UBDs) have been described in several endosomal sorting complexes required for transport (ESCRT). Using available structural information, we have investigated the role of the multiple UBDs within ESCRTs during MVB cargo selection. We found a novel UBD within ESCRT-I and show that it contributes to MVB sorting in concert with the known UBDs within the ESCRT complexes. These experiments reveal an unexpected level of coordination among the ESCRT UBDs, suggesting that they collectively recognize a diverse set of cargo rather than act sequentially at discrete steps
Modification of Drosophila p53 by SUMO Modulates Its Transactivation and Pro-apoptotic Functions*S⃞
Conjugation to SUMO is a reversible post-translational modification that
regulates several transcription factors involved in cell proliferation,
differentiation, and disease. The p53 tumor suppressor can be modified by
SUMO-1 in mammalian cells, but the functional consequences of this
modification are unclear. Here, we demonstrate that the Drosophila
homolog of human p53 can be efficiently sumoylated in insect cells. We
identify two lysine residues involved in SUMO attachment, one at the C
terminus, between the DNA binding and oligomerization domains, and one at the
N terminus of the protein. We find that sumoylation helps recruit
Drosophila p53 to nuclear dot-like structures that can be marked by
human PML and the Drosophila homologue of Daxx. We demonstrate that
mutation of both sumoylation sites dramatically reduces the transcriptional
activity of p53 and its ability to induce apoptosis in transgenic flies,
providing in vivo evidence that sumoylation is critical for
Drosophila p53 function
The HECT Domain of the Ubiquitin Ligase Rsp5 Contributes to Substrate Recognition*
Ubiquitin modification of endosomal membrane proteins is a signal for active inclusion into the Multivesicular Body (MVB) pathway, resulting in lysosomal degradation. However, the endosome represents a dynamic site of protein sorting with a majority of proteins destined for recycling, rather than MVB targeting. Substrate recognition by ubiquitin ligases is therefore highly regulated. We have investigated substrate recognition by the Nedd4 ortholog Rsp5 as a model for understanding ligase-substrate interactions. Rsp5 interacts directly with its substrate Cps1 via a novel interaction mode. Perturbation of this mode of interaction revealed a compensatory role for the Rsp5 adaptor Bsd2. These results highlight the ability of Rsp5 to interact with substrates via multiple modalities, suggesting additional mechanisms of regulating this interaction and relevant outcomes
